Leur \u00e9tat d&#8217;ionisation d\u00e9pend du pH de la solution.<\/p><p>Les prot\u00e9ines sont des polym\u00e8res d&#8217;acides amin\u00e9s. La liaison peptidique unit deux acides amin\u00e9s selon une g\u00e9om\u00e9trie qui conditionne les structures d\u2019ordre sup\u00e9rieur.<\/p><p>Les propri\u00e9t\u00e9s physico-chimiques de la liaison peptidique et des radicaux des acides amin\u00e9s permettent aux prot\u00e9ines d\u2019acqu\u00e9rir une structure tridimensionnelle secondaire, tertiaire et quaternaire.<\/p><p>La structure d\u2019une prot\u00e9ine peut \u00eatre \u00e9tudi\u00e9e par des m\u00e9thodes physico-chimiques.<\/p><p>La fonction d\u2019une prot\u00e9ine d\u00e9pend de son affinit\u00e9 et de sa sp\u00e9cificit\u00e9 pour un ligand au niveau d\u2019un site d\u2019interaction. L\u2019affinit\u00e9 et la sp\u00e9cificit\u00e9 d\u2019un site d\u2019interaction sont li\u00e9es \u00e0 sa structure tridimensionnelle et \u00e0 la nature des acides amin\u00e9s constitutifs.<\/p><p>La s\u00e9quence en acides amin\u00e9s et la structure tridimensionnelle des prot\u00e9ines peuvent leur conf\u00e9rer des propri\u00e9t\u00e9s m\u00e9caniques.<\/p><p>Les macromol\u00e9cules prot\u00e9iques sont des structures dynamiques du fait de la labilit\u00e9 des interactions faibles, ce qui participe \u00e0 leur fonction.<\/p><p>La coop\u00e9rativit\u00e9 est permise par les changements conformationnels des prot\u00e9ines (allost\u00e9rie).<\/p><p>Certaines prot\u00e9ines peuvent subir des modifications post-traductionnelles (glycosylation, phosphorylation).<\/p><p>Les connaissances sur l\u2019affinit\u00e9 et la sp\u00e9cificit\u00e9 des interactions prot\u00e9ine-ligand ont permis de mettre au point des techniques de purification et d\u2019en d\u2019\u00e9valuer l\u2019efficacit\u00e9.
